Structural Biochemistry
本课程从分子水平研究生物大分子的三维结构及其与功能的关系。内容涵盖蛋白质的氨基酸组成、一级结构测定、二级结构类型(α-螺旋、β-折叠)、三级结构与结构域、四级结构与亚基组装,以及核酸的双螺旋结构、超螺旋与染色体结构。重点讲授蛋白质折叠机制、分子伴侣、构象变化与功能调节,介绍X射线晶体学、核磁共振、冷冻电镜等结构生物学研究方法。
This course studies the three-dimensional structures of biological macromolecules and their relationship to function at the molecular level. Topics include protein amino acid composition, primary structure determination, secondary structure types (α-helix, β-sheet), tertiary structure and domains, quaternary structure and subunit assembly, as well as DNA double helix, supercoiling, and chromosome structure. Emphasis on protein folding mechanisms, chaperones, conformational changes, and functional regulation, with introduction to X-ray crystallography, NMR, and cryo-EM methods.
