本课程以实验操作为主,系统训练分子生物学常用实验技术。内容包括核酸提取与纯化、PCR扩增技术、琼脂糖凝胶电泳、限制性酶切与DNA连接、感受态细胞制备与转化、质粒提取、实时荧光定量PCR、蛋白质表达与纯化、Western Blot等。课程强调实验原理与操作技能并重,培养学生独立完成分子生物学实验的能力及分析解决实验问题的能力。

This course focuses on laboratory training, systematically teaching common molecular biology techniques. Topics include nucleic acid extraction and purification, PCR amplification, agarose gel electrophoresis, restriction digestion and DNA ligation, competent cell preparation and transformation, plasmid extraction, real-time qPCR, protein expression and purification, and Western Blot.

作业标题:PCR扩增特定基因及序列验证实验报告

从细胞样本中提取基因组DNA,设计特异性引物,通过PCR扩增目的基因片段。进行琼脂糖凝胶电泳检测扩增产物,胶回收纯化后连接到T载体,转化大肠杆菌感受态细胞,挑取阳性克隆进行菌液PCR验证及测序。提交完整的实验报告,包括实验原始记录、结果图片、序列比对分析及讨论。

Extract genomic DNA from cell samples, design specific primers, amplify the target gene fragment by PCR. Perform agarose gel electrophoresis, gel extraction, ligation into T-vector, transformation into E. coli competent cells, colony PCR verification, and sequencing.
